TLDR
Develop PALMS-based probes and proteomics workflows to map compound binding sites on target proteins in native cellular systems.
General Summary:
The candidate will contribute to the development of a PALMS-based method to identify the binding site of a compound on its target protein in a cellular context. While the PALMS workflow is currently applicable to purified recombinant proteins, its implementation for transmembrane proteins such as GPCRs and ion channels remains challenging. Extending PALMS approach to native cellular systems would represent a significant advancement.
Working closely with the Mass Spectrometry, Biochemistry, and Bioinformatics teams, the candidate will participate in the development of novel probes enabling the identification of binding sites on proteins of interest in a cellular context. Various biochemical approaches will first be employed and validated before analysis by mass spectrometry to determine the peptide sequence and the specific amino acid involved in compound binding.
Missions and responsibilities:
- Review, understand and summarize scientific literature.
- Contribute to proteomic studies to identify the binding site of compounds on known targets using both recombinant protein models and cellular systems.
- Identify and implement innovative biochemical approaches compatible with proteomics workflows, based on literature findings and in collaboration with the supervisor.
- Interpret and exploit experimental results within their biological context, in coordination with Biochemistry, Proteomics, and Bioinformatics teams.
- Participate in the development and implementation of new methodologies and strategies for proteomics sample preparation.
- Communicate effectively through written reports, oral presentations, and scientific discussions.
- Demonstrate strong organizational, multitasking, analytical, and communication skills, with the ability to clearly present scientific findings to project teams.
- Apply a proactive and solution-oriented approach to problem-solving.
- Collaborate closely with members of the different teams involved and cross-functional partners.
Knowledge, skills and abilities:
- Excellent communication and interpersonal skills.
- Hands-on experience with cell culture, including cell lines, primary cells, bacteria, and/or yeast.
- Knowledge in proteomics sample preparation for mass spectrometry analysis.
- Experience with SDS-PAGE and Western blotting techniques.
- Strong scientific rigor and excellent analytical capabilities.
- Ability to work effectively within multidisciplinary and collaborative teams.
Experience and education:
- Master's degree/engineering student with a background in Chemistry, Biochemistry, Proteomics
